Thymosin alpha-1 is a synthetic peptide of 28 amino acids, corresponding to the N-terminal fragment of prothymosin alpha. Its sequence begins with acetylation at the N-terminus, a modification that affects stability and receptor interaction. The peptide is acidic, with a calculated isoelectric point near 4.2, and carries no disulfide bonds, so its secondary structure is largely flexible in solution. Molecular mass is approximately 3108 daltons. The native form was first isolated from bovine thymus tissue, while pharmaceutical material is produced by solid-phase peptide synthesis.
Within the immune system, the peptide acts on several cell types rather than a single target. Reported activities include promotion of T-cell maturation, enhancement of natural killer cell activity, and modulation of cytokine production by dendritic cells and macrophages. Some of these effects appear to operate through toll-like receptor signaling, though the precise receptor-level mechanism remains debated. Whether the observed immune changes translate into clinical benefit is a separate question and depends on the indication studied.
The peptide was described in the 1970s as a component of thymic extracts, and early research focused on restoring immune function in immunodeficiency states. A synthetic version entered clinical development in the 1980s and is approved as a drug in several countries for conditions such as chronic hepatitis B and certain immunodeficiencies. Approval status varies widely by jurisdiction, and in the United States it is not an approved therapeutic. Regulatory and clinical positions differ, so statements about efficacy should be tied to specific indications and studies.
Like most short peptides, thymosin alpha-1 is susceptible to hydrolysis under strongly acidic or basic conditions and to oxidation when exposed to air over long periods. The acetylated amino terminus blocks one common degradation route, which contributes to the molecule's relative robustness in solution. Lyophilized material generally retains potency for extended periods when kept cold and dry. Once reconstituted, aqueous solutions are less stable and are typically used within a defined window rather than held indefinitely at ambient temperature.
Routine handling calls for storage of the lyophilized powder at refrigerated temperatures, away from light, in a sealed container. Working solutions are often prepared in sterile water or buffer and kept cold between uses. Repeated freeze-thaw cycles are generally avoided because they can promote aggregation and loss of material. Laboratories usually record lot number, reconstitution date, and storage conditions so that any change in behavior can be traced to a specific preparation.
Reverse-phase high-performance liquid chromatography is the standard technique for assessing purity and concentration, because the peptide's hydrophobicity allows clean separation from related impurities. Mass spectrometry confirms molecular identity and detects sequence errors or truncations. Amino acid analysis and peptide mapping supply additional structural confirmation when required. Chromatographic purity values reported on certificates of analysis describe the proportion of the main peak and do not by themselves establish biological activity.

Thymosin alpha 1 was identified in 1977 as a component of thymosin fraction 5, a heterogeneous preparation used in early studies of thymic function. Investigators purified the active material and determined its amino acid sequence, which enabled chemical synthesis. Work in the following decades concentrated on T-cell maturation and immune reconstitution in animals and small human cohorts. Early preparations varied in composition, so results from that period are difficult to compare with studies using defined synthetic peptide.
Clinical research has examined the peptide in chronic hepatitis B and C, as a vaccine adjuvant, and in sepsis and oncology settings. Findings across trials are mixed; some report changes in selected immune markers, while others find no clear clinical benefit. Many studies are small and define outcomes differently, which limits comparison. Regulatory approval is confined to a few countries, and the compound is not an approved drug in the United States or most of Europe.

== History == Sauk City restaurateurs and husband and wife team George and Ruth Culver started their fast food careers as the owners of an A&W on Phillips Boulevard (U.S. Highway 12) in 1961. In 1968, they purchased a resort-styled restaurant at Devil's Lake called The Farm Kitchen. Their son, Craig Culver, worked for a local McDonald's right out of college in 1973. George, Ruth, their son Craig, and his wife Lea opened the first Culver's Frozen Custard and ButterBurgers in Sauk City, Wisconsin on July 18, 1984. Craig was CEO of Culver's from its inception until mid-2015.
According to the author of the study there were 137 such events in Thailand during this period. Other ASEAN nations ranked in the study were Myanmar, ranked 3; Philippines, 5; and Vietnam, 8. Climate Change Performance Index (CCPI) 2017: Of 58 nations responsible for about 90% of global energy-related CO2 emissions, Thailand was ranked 42d in taking "action on climate protection" (1=best, 61=worst), earning an assessment of "poor". Other ASEAN nations ranked were Indonesia, 22 "good"; Malaysia, 44, "poor"; and Singapore, 54, "very poor". Global Climate Risk Index 2017: Thailand ranked number 10 of 180 nations most affected by "...weather-related loss events..." during the period 1996-2015 (1=most affected, 180=least affected) according to Germanwatch. Other ASEAN nations ranked in the top 10 were Myanmar, 2; Philippines, 5; and Vietnam, 8. Climate Central: Thailand is ranked seventh (higher number=lower risk) on the "Top 20 Most At-Risk Countries" for exposure to sea level rise and coastal flooding. It estimates the number of "People living on land that will be below sea level or chronic flood levels by the end of the century, assuming current emissions trends continue, and medium sensitivity of sea level to warming." In Thailand, eight million persons, or 12% of the population, are "at-risk" according to the study. Six of ASEAN's ten member-states are on the at-risk list. Climate Change Performance Index 2016: Thailand ranked 49 of 61 (1=best, 61=worst) countries, putting it in the "Very Poor" performance category.

=== In media === In the Stephen King short story collection Night Shift, the final story of the book, "The Woman in the Room", tells a tale in which the main character contemplates and then finally performs a mercy killing using a drug called "Darvon Complex". In his 1976 song "My Head Hurts My Feet Stink and I Don't Love Jesus" Jimmy Buffett references taking a Darvon to relieve his hangover.
These reviews suggest that lisdexamfetamine is persistently effective at treating BED and is associated with significant reductions in the number of binge eating days and binge eating episodes per week. Furthermore, a meta-analytic systematic review highlighted an open-label, 12-month extension safety and tolerability study that reported lisdexamfetamine remained effective at reducing the number of binge eating days for the duration of the study. In addition, both a review and a meta-analytic systematic review found lisdexamfetamine to be superior to placebo in several secondary outcome measures, including persistent binge eating cessation, reduction of obsessive-compulsive related binge eating symptoms, reduction of body-weight, and reduction of triglycerides. Lisdexamfetamine, like all pharmaceutical amphetamines, has direct appetite suppressant effects that may be therapeutically useful in both BED and its comorbidities. Based on reviews of neuroimaging studies involving BED-diagnosed participants, therapeutic neuroplasticity in dopaminergic and noradrenergic pathways from long-term use of lisdexamfetamine may be implicated in lasting improvements in the regulation of eating behaviors that are observed.
==== Science and Technology Center ==== The Science and Technology Center (S/T) is a highly challenging four-year curriculum which provides college-level academic experiences in science, mathematics, and technology. The program is not a true magnet program, as students are admitted into the S/T program based on competitive examination only, as opposed to the standard magnet lottery process. Of twenty-eight possible credits, a student is required to obtain a minimum of thirteen credits in specific mathematics, pre-engineering technology, research and science courses. In grades nine and ten, the program consists of common experiences courses for all student. In grades eleven and twelve, each student must choose course work from at least one of four major study areas. Students are expected to be enrolled in a full schedule of classes during the entire four-year program. External experiences are possible and encouraged, but must be a direct extension or enrichment of the Science and Technology Program, and have the recommendation of the Science and Technology Center Coordinator prior to approval by the principal. The program is offered at three centers — Eleanor Roosevelt High School in northern Prince George's County, Oxon Hill High School in southern Prince George's County, and Charles Herbert Flowers High School in central Prince George's County. Students attend the center that serves their legal residence. Transportation is provided for all students. Each school is a four-year comprehensive high school, as well as a Science and Technology Center.

It corresponds to a fragment of the larger protein prothymosin alpha, which is present in many tissues. The isolated 28-amino-acid peptide was originally obtained from thymus preparations, and the pharmaceutical product is synthesized rather than extracted. The term therefore describes both a natural fragment and a manufactured drug substance.
Thymosin alpha-1 is a single defined 28-residue peptide, while the broader family includes unrelated peptides such as thymosin beta-4. The shared name reflects historical isolation from thymus tissue rather than a common structure. Confusion between the two is common in older literature.
No single pathway fully accounts for its reported effects. Several studies describe interaction with innate immune receptors and downstream cytokine changes, but the complete picture is not settled. Open questions remain about which effects occur at physiological concentrations.
The lyophilized solid is normally held at 2 to 8 °C in a sealed, light-protected container. Dry storage limits both hydrolysis and microbial growth. Material kept this way remains stable for the shelf life stated by the supplier.
